It is used for the treatment of idiopathic pulmonary fibrosis (IPF) and along with other medications for some types of non-small-cell lung cancer.
Mechanism of action:
Ofev (Nintedanib ) is a small molecule that targets multiple receptor tyrosine kinases (RTKs) and non-receptor tyrosine kinases (nRTKs). Ofev (Nintedanib ) inhibits the following RTKs: platelet-derived growth factor receptor (PDGFR) α and β, fibroblast growth factor receptor (FGFR) 1-3, vascular endothelial growth factor receptor (VEGFR) 1-3, and Fms-like tyrosine kinase-3 (FLT3). Among them, FGFR, PDGFR, and VEGFR have been implicated in IPF pathogenesis. Ofev (Nintedanib ) binds competitively to the adenosine triphosphate (ATP) binding pocket of these receptors and blocks the intracellular signaling which is crucial for the proliferation, migration, and transformation of fibroblasts representing essential mechanisms of the IPF pathology. In addition, Ofev (Nintedanib ) inhibits the following nRTKs: Lck, Lyn and Src kinases. The contribution of FLT3 and nRTK inhibition to IPF efficacy is unknown.
Dosage & Administration:
Use Ofev (Nintedanib ) as directed by your doctor. Check the label on the medicine for exact dosing instructions.· An extra patient leaflet is available with Ofev (Nintedanib ). Talk to your pharmacist if you have questions about this information.
· Take Ofev (Nintedanib ) by mouth with food and liquid.
· Swallow Ofev (Nintedanib ) whole. Do not break, crush, or chew before swallowing.
· Take Ofev (Nintedanib ) on a regular schedule to get the most benefit from it.
· If you miss a dose of Ofev (Nintedanib ), skip the missed dose and go back to your regular dosing schedule. Do not take 2 doses at once.
Ask your health care provider any questions you may have about how to use Ofev (Nintedanib ).
Diarrhea, nausea, vomiting, abdominal pain, decreased appetite, and weight loss may occur. If any of these effects persist or worsen, tell your doctor or pharmacist promptly. Diarrhea, nausea, and vomiting can be persistent and/or severe. Your doctor may have you drink more fluids if these symptoms occur and prescribe medications to treat the diarrhea, nausea, and vomiting.
Before taking Ofev (Nintedanib ), tell your doctor or pharmacist if you are allergic to it; or if you have any other allergies. This product may contain inactive ingredients which can cause allergic reactions or other problems. Talk to your pharmacist for more details.
Store Ofev (Nintedanib ) at room temperature, between 68 and 77 degrees F (20 and 25 degrees C). Store away from heat, moisture, and light. Keep Ofev (Nintedanib ) out of the reach of children and away from pets.
